Dr. Ciara L. Freeman is looking for a motivated Postdoctoral Fellow to lead a high-impact project on next-generation CAR-T cell therapy for relapsed or refractory multiple myeloma.

The ideal candidate will possess strong skills in flow cytometry, cellular immunology, and molecular biology, with the ability to design and execute complex experiments. While prior CAR-T manufacturing experience is a plus, skilled scientists without it are encouraged to apply, as comprehensive training will be provided.

Position Highlights:
  • Work in a world-class NCI-designated Comprehensive Cancer Center with access to state-of-the-art core facilities, animal modeling, and GMP manufacturing.
  • Collaborate with a multidisciplinary team of physician-scientists, immunologists, and translational researchers.
  • Present findings at major scientific conferences and publish in leading journals.
  • Contribute directly to the development of next-generation CAR-T therapies for patients with advanced cancers.

The Ideal Candidate:
  • Must have extensive, hands-on expertise in multi-color flow cytometry - including panel design, troubleshooting, sample preparation, data acquisition, and high-dimensional analysis.
  • Strong technical skills in molecular cloning, mammalian cell culture, and immune functional assays.
  • Experience with in vivo cancer models.
  • Demonstrated ability to design and execute complex experimental workflows, analyze data rigorously.

Preferred (Can Be Learned On-Site):
    • Experience with CAR-T or other engineered immune cell manufacturing workflows.
    • Experience with antibody engineering platforms.
    • Familiarity with regulatory processes for preclinical cellular therapy development.

Responsibilities:

1. CAR Construct Development & Optimization
  • Lead the molecular design and engineering of CAR constructs using novel antigen-binding domains and optimized intracellular signaling architectures.
  • Evaluate and refine construct designs to improve anti-tumor potency, persistence, and safety.

2. In Vitro Functional Characterization
  • Perform extensive multi-color flow cytometry for immune phenotyping, functional assessment, and CAR expression analysis.
  • Execute cytotoxicity, cytokine release, proliferation, and activation assays using tumor cell lines and primary patient samples.
  • Conduct long-term functionality and metabolic profiling assays.

3. In Vivo Efficacy & Safety Studies
  • Design and perform murine models to evaluate tumor clearance, persistence, and biodistribution.
  • Monitor tumor burden and immune cell dynamics using imaging and flow cytometry-based biodistribution analysis.
  • Conduct off-target effect assessments and histopathological analyses.

4. Translational & Regulatory Support
  • Generate rigorous preclinical datasets to support the advancement of CAR-T candidates toward clinical evaluation.
  • Collaborate with GMP manufacturing teams, core facilities, and regulatory specialists to ensure translational readiness.

Credentials and Qualifications:
  • MD or PhD in Immunology, Cancer Biology, Molecular Biology, or a related biomedical discipline.
  • Preference will be given to candidates with a minimum of 2 years of postdoctoral or equivalent advanced research experience in cellular immunology, cancer immunotherapy, or related biomedical research.
  • Should be published in peer-reviewed journals.
